Bifenthrin is een van de meest gebruikte kunstmatige pyrethroïde pesticiden. Het richt zich op het zenuwstelsel van insecten, voornamelijk op natriumkanalen in zenuwcelmembranen. Het overmatige gebruik van bifrenthrin kan leiden tot een toename van de resistentie tegen plaaginsecten. Bovendien zijn er slechts enkele onderzoeken die de cytotoxische beweging ervan beschrijven.
Er was een reeks bioassays uitgevoerd en de resultaten bevestigden dat bifenthrin een grote vaardigheid heeft om DNA-beschadiging en de remming van de levensvatbaarheid in Spodoptera frugiperda (Sf9)-cellen te induceren . Monodansylcadaverine-kleuring en transmissie-elektronenmicroscoopassays waren gebruikt om belangrijke reeksen van autofagosomen en mitochondriale disfunctie in het cytoplasma te bekijken.
Bovendien bevestigde Western-blot-evaluatie een opwaartse regulatie in LC3-II- en beclin-1-eiwitexpressie en een neerwaartse regulatie in p62-expressie , die bijdroeg aan de cytotoxische impact van bifenthrin op Sf9-cellen. Algemeen, bifenthrin heeft een aanzienlijke invloed op de levensvatbaarheid van Sf9-cellen door DNA-beschadiging en autofagie te induceren. Een ernstige belemmering voor het vinden van membraaneiwitten door middel van biofysische strategieën is de kwestie van het produceren van voldoende hoeveelheden voorraden voor nuttig en structureel onderzoek. Om het beoogde membraaneiwit heteroloog, in het bijzonder een eukaryoot eiwit, tot overexpressie te brengen , is een belangrijke stap het zoeken naar het optimale gastheerexpressiesysteem en het uitvoeren van daaropvolgende expressieoptimalisatie.
In dit hoofdstuk beschrijven we protocollen voor het screenen van membraaneiwitproductie met behulp van bacteriële en insectencellen, solubilisatiescreening, grootschalige productie en algemeen gebruikte zuiveringsstrategieën voor affiniteitschromatografie. We richten ons op veelvoorkomende optimalisatiesituaties, gelijk aan promotors en tags, en beschrijven de huidige strategieën die in elk laboratorium kunnen worden gebruikt zonder gespecialiseerde dure uitrusting.
Met name voor insectencellen zijn GFP-fusies met name nuttig voor lokalisatie en in-gel fluorescentiedetectie van de eiwitten op SDS-PAGE . We geven gedetailleerde protocollen die kunnen worden gebruikt om de meest effectieve expressie- en zuiveringssituaties voor membraaneiwitonderzoek weer te geven.
Bindingsspecificiteit van ostreolysin A6 richting Sf9 insectencellen cel lipiden.
Van oesterzwammen (Pleurotus spp.) is de laatste tijd bewezen dat ze insecticide bi-component eiwitcomplexen leveren die voornamelijk gebaseerd zijn op de aegerolysine-eiwitten. Een taak voor deze eiwitten is dus aangewezen voor de verdediging en veiligheid van de paddenstoel, en we stellen voor om ze te gebruiken als nieuwe milieuvriendelijke bio-insecticiden.
Deze op aegerolysine gebaseerde eiwitcomplexen permeabiliseren synthetische lipideblaasjes door middel van aegerolysinebinding aan een insect-specifiek sfingolipide, ceramidefosfoethanolamine (CPE), en ze zijn cytotoxisch voor de Spodoptera frugiferda (Sf9) insectencellijn.
Tandem-massaspectrometrie-evaluatie van de Sf9-lipidoom-ontdekte lipiden die niet vooraf in de literatuur zijn vermeld, samen met specifiek C14-sfingosine-gebaseerde CPE-moleculaire soorten, die ~ vier mol% van het gehele lipidoom omvatten.
Aanvullende evaluatie van de lipidenbindingsspecificiteit van een aegerolysine van P. ostreatus, ostreolysine A6 (OlyA6), tot lipidevesikels die zijn samengesteld uit economische lipiden, tot lipidevesicles die zijn samengesteld uit het volledige lipide-extract van Sf9-cellen, en tot HPLC-gescheiden Sf9-cellipide fracties die ceramiden bevatten, bevestigden CPE als de belangrijkste OlyA6-receptor, maar benadrukten daarnaast het belang van membraan-ldl-cholesterol voor de vorming van stevige en veilige interacties van OlyA6 met synthetische en zuivere lipidemembranen.
Bindingstests uitgevoerd met glycaanarrays en vloerplasmonresonantie, waaronder ongewervelde-specifieke glycanen, sloten deze sacchariden uit als potentiële extra OlyA6-receptoren.
Koel-actieve extracellulaire lipase: Expressie in Sf9 insect cellen , zuivering en katalyse.
Chilly-actieve lipasen krijgen tegenwoordig bijzondere aandacht, omdat ze meer en meer worden gebruikt in tal van industrieën die gelijkwaardig zijn aan hoogwaardige chemische synthese, maaltijdverwerking en wasmiddel. Binnen het huidige onderzoek werd een extracellulair lipase-gen van Yarrowia lipolytica (LIPY8) gekloond en tot expressie gebracht door het baculovirus-expressiesysteem.
Het recombinante lipase (LipY8p) werd gezuiverd met behulp van chromatografische strategieën, wat leidde tot een zuiveringsprobleem van 25,7-voudig met een specifieke inspanning van 1102,9 U/mg voor olijfolie. De voor de hand liggende molecuulmassa van gezuiverd LipY8p was 40 kDa. Het enzym was het meest energiek bij pH 7,5 en 17 °C . Een correcte en snelle prognose van griep en infectie is belangrijk om een vroegtijdige antivirale behandeling mogelijk te maken en de sterfte als gevolg van seizoens- en epidemische infecties terug te dringen. Immunochromatografie is een van de meest typische strategieën die worden gebruikt voor de prognose van seizoensgriep bij de mens; desalniettemin is het veel minder efficiënt bij het diagnosticeren van het pandemische influenzavirus.
Op dit moment zijn snelle diagnostische kits voor pandemisch influenzavirus afhankelijk van de detectie van nucleoproteïne (NP) of hemagglutinine (HA). NP-detectie onthult een grotere specificiteit en is extra delicaat dan HA-detectie. Bij dit onderzoek hebben we tijdsafhankelijke expressiesituaties gescreend en hierin rapporteren we optimale situaties voor de expressie van recombinant nucleoproteïne (rNP), die 48 uur na een infectie was.
Daarnaast rapporteren we met behulp van de uitgedrukte rNP in een snelle diagnose van griep (SGT i-flex Influenza A&B Check). We construeerden expressievectoren die rNP (antigeen) van influenza A en B in insectencellen (Sf9-cellen) synthetiseerden, gebruikten het gezuiverde rNP voor de immunoassay, bekijk de verpakking en onderscheidden duidelijk NP’s van influenza A en influenza B met behulp van deze snelle influenza diagnostisch pakket. Deze strategie zou het geval kunnen verbeteren van een snelle kijk op kits voor griep die NP gebruiken.
Antitumor uitoefening van Escherichia coli Shiga-toxine A subeenheid geleverd door SF9 insectencellen cellen .
De meeste vormen van kanker blijven over de hele wereld een ernstig nadeel voor het welzijn. Een Shiga-toxine is een bacterieel toxine dat gewoonlijk wordt geproduceerd door Shigella dysenteriae en Escherichia coli. Een subeenheid van het Shiga-toxine (StxA) is een cytotoxisch middel dat zou kunnen worden gebruikt om de dood in de meeste kankercellen te induceren. StxA tot expressie gebracht vanuit baculovirus werd geëvalueerd in een pTriExTM-expressievector. De baculovirusvector werd gebruikt voor de A-subeenheidtoevoer van StxA. De cytotoxiciteit van StxA-cellen werd door het virus geïnduceerd en beoordeeld binnen de MCF7- en HeLa-celstammen.
Evenals de cytotoxiciteit van de meeste kankers van de tot expressie gebrachte StxA werd bovendien beoordeeld in de meeste kankers die bij muizen werden geïnduceerd. De cytotoxiciteit van het recombinante StxA-baculovirus met totaal verschillende multipliciteiten van een infectie (MOI) werd gemeten.
De resultaten bevestigden dat belangrijke cytotoxiciteit kan worden geïnduceerd op het borstepitheel van zoogdieren, de meeste kankercelstammen, MCF7- en HeLa-cellen met een MOI ≥ 2. De resultaten bevestigden bovendien {dat een} kwaadaardige tumor geïnduceerd door MCF7 zou kunnen worden geremd in een mannequin met de meeste kankers . Vanwege dit feit kan worden geconcludeerd dat StxA, tot expressie gebracht door baculovirus, kan worden gebruikt voor in vitro en in vivo genlevering.
